If the color of a cell will not change, make sure it is not protected.

Cannot change cell color on a particular cell

if a cell has conditional formatting, any other color formatting you
try to do will be overridden by the conditional.

say A1 is formatted (conditionally) to be white when empty, and yellow
when it contains a value.

if you've got "yes" in A1, you can try to format it any color you
want, but you'll only get the yellow as set above.

maybe this helps?
